The most important characteristic of ESCs is their pluripotency, or ability to differentiate into all three primitive cell lineages in the body -- endoderm, ectoderm and mesoderm. In other words, they can develop into each of the more than 200 cell types of the adult body when given sufficient and necessary stimulation for a specific cell type. Researchers working with hESCs must continually screen them to ascertain pluripotency. The TrulyStem kit is a robust and sensitive multi marker quantitative real-time PCR method, with which you can measure 13 genes regulated upon differentiation. The relative differentiation state of the cells can be determined by calculating a collective expression index based on the mRNA levels of the genes from your cells and the provided cDNA controls.

Materials included

• Primer solutions for 13 stem cell differentiation markers. 100 rxns.

• Positive Control cDNA from non-differentiated stem cells, cell lineage AS002. 13 rxns, one reaction for each gene.

• Negative Control cDNA from differentiated stem cells, cell lineage AS002. 13 rxns, one reaction for each gene.

• The addition of real-time PCR regents is necessary. Primer sets are validated for use with most commercial available master mixes for qPCR containing SYBR Green I.
