Digital PCR is an exceptionally accurate and sensitive method to detect and count individual nucleic acid molecules. Digital PCR is based on the concept that PCR produces a very large amount of product even from a single template molecule. In order to determine the number of molecules in a sample with high precision, the sample is divided into a large number of aliquots/reactions that are analyzed separately by traditional end-point PCR. Some of these reactions contain the target molecule (positive) while others do not (negative). Hence, by calculating the number of positive PCR’s, virtually the number of template molecules that were present initially in the sample is counted, without reference to standards or endogenous controls.
- High precision measurements
- Rare mutation detection
- Copy number variation
- Calibration of standards
- Quantification of NGS libraries
- Analysis of inhibitory samples
- Splice variant preference
- Allelic preference
- Cis vs trans localization
TATAA Biocenter researchers were among the first exploiting dPCR and published one of the first papers using a high throughput platform pioneering the field of subcellular expression profiling: Intracellular expression profiles measured by real-time PCR tomography in the Xenopus laevis oocyte, Nucleic acids research 36 (2), 387-392 (2008). TATAA Biocenter researchers also coauthored the dPCR MIQE guidelines: The digital MIQE guidelines: minimum information for publication of quantitative digital PCR experiments. Clinical chemistry 59 (6), 892-902 (2013).
At TATAA we have all the leading dPCR platforms always choosing the most appropriate for purpose.
